What is NSI 189 Phosphate?

NSI 189 phosphate is a novel neurogenic compound that acquires pleiotropic properties.

Emerging evidence also suggests NSI 189 phosphate powder has regionally specific effects, with neurogenesis occurring primarily in the hippocampus’s subventricular zone.

According to preliminary findings from a proof-of-concept study, NSI 189 may be capable of alleviating depressive symptoms and improving cognitive function in adults with DSM-5-defined Major Depressive Disorder (MDD).

Possible Applications of NSI 189 Phosphate

NSI 189 Phosphate and Cognitive Impairment

Phase I and II clinical trials that examined depression also identified possible advantages for cognition.

NSI 189 Phosphate and Major Depressive Disorder (MDD)

Twenty-four MDD patients were recruited, and their severity and conditions prior to recruitment were confirmed via remote interviews. Eligible patients were given NSI-189 or a placebo for 28 days in an inpatient setting, with safety, pharmacokinetics (PK), and efficacy evaluated. Outpatient follow-up visits continued until day 84. NSI-189 was well tolerated at all doses, with no severe side effects.

NSI 189 Phosphate and Ischemic Stroke

In adult Sprague-Dawley rats, oral administration of NSI-189 beginning at 6 hours after middle cerebral artery occlusion and continuing every day for the following 12 weeks led to a significant improvement in stroke-induced motor and neurological deficits, which was maintained up to 24 weeks post-stroke.

NSI 189 Phosphate and Diabetic Neuropathy

Oral administration of NSI-189 to male and female murine models of type 1 and type 2 diabetes prevented numerous functional and structural indicators of small and large fiber peripheral neuropathy, increased hippocampal neurogenesis, synaptic markers, volume, and safeguarded long-term memory.

NSI-189’s mechanism of action is not fully understood. According to laboratory research, this experimental compound may work by promoting neurogenesis or the formation of new nerve and brain cells.
